This is Sentinels of The Multiverse Enhanced Card Game. Sentinels of the Multiverse is extremely popular at this point, with a legion of fans. At it's simplest, Sentinels of the Multiverse is a cooperative fixed-deck card game that's dressed in comic book attire.
It's a large and in-depth game. The players play as one of ten heroes against one of the four villains and there are four dynamic environments to battle in. This is the Enhanced version of Sentinels of the Multiverse. It's the second printing and contains more room in the box to accomadate the expansion cards of Rook City and Infernal Relics. Also, it comes with 36 divider cards. Super cool.
